1. Introduction
The importance of dNTP quality in today’s molecular biology is often overlooked. Bioline’s ultra pure dNTPs are enzymatically manufactured to maximise purity and quality. Our dNTPs will work even under the most difficult and sensitive situations, such as long-distance & qPCR, Reverse Transcription and DHPLC.
2. Performance and Sensitivity
Bioline dNTPs meet exacting standards and are suitable for the most demanding applications such as long distance PCR and DNA synthesis, Real-Time PCR and Reverse Transcription. Bioline dNTPs can be employed in highly sensitive assays such as low-copy or rare-message assays.
3. Purity
Bioline dNTPs are enzymatically synthesised from dNMPs by enzymatic phosphorylation. This process eliminates impurities and PCR inhibitors such as modified nucleotides, inorganic pyrophosphates and tetraphosphates. The dNTPs are purified with preparative HPLC and possess at least 99% purity.
DNA fragments amplified using Bioline dNTPs have been assayed for use on DHPLC. dNTP purity is a critical consideration for DHPLC usage, as there must be no contamination by monophosphates or incorrectly synthesised/ modified bases. Contaminating nucleotides or derivatives may cause premature strand termination and result in unwanted peaks.
4. Quality Control
dNTPs are tested for the absence of DNase, RNase, Protease and nicking activity. Tests to confirm the absence of human and bacterial DNA are also carried out. Each batch is validated rigorously for purity by HPLC, and tested for performance in a wide range of PCR templates, including a long-distance 20kb functional test.
5. Configuration
Bioline dNTPs are available as convenient 100 mM sets or as 10, 40 and 100 mM ready-to-go mixes, which are designed to save time and reduce the risk of contamination. The dNTP solutions are ready-to-use at pH 7.0 in Lithium salts.
6. Extended Storage Life
Bioline dNTPs have the distinct advantage of being presented as Lithium Salts as opposed to Sodium Salts. Lithium presentation makes dNTPs more resistant to repeated freeze/ thawing. A Lithium salt solution is also an unsuitable environment for bacteria and fungi, so the dNTPs remain sterile throughout the storage and usage period. Bioline dNTPs are stable for 12 months when stored in a -20ºC constant-temperature freezer.

11. dUTP Mix & ACGU + T Mix
- Used in conjunction with UNG to prevent carry-over of amplified products into subsequent amplifications.
- Convenient pre-optimised, premixed PCR grade solutions containing dATP, dCTP, dGTP and dUTP, as well as dTTP in the case of ACGU + T Mix.
- Applications: Real-Time PCR.
Bioline dUTP Mix and ACGU + T Mix enable users to employ the efficient strategy of incorporating dUTP into amplification reactions. In this process, dUTP replaces dTTP in amplified products. Before a subsequent DNA amplification is commenced, treatment with Uracil DNA Glycosylase (UNG) will result in the selective cleavage of the Uracil base in the dUTP-containing DNA, therefore preventing carry-over contaminations.
Note: dUTP Mix is the standard preparation recommended. However, please use ACGU + T Mix for >500 bp amplifications.
